Methods are providedfor using methotrexate (MTX) activeagents in which reduced host toxicity isobserved. Aspects of the methodsin-clude administering to a subject anef-fective amount of an MTX active agentin conjunction with a MTXtoxicity-re-ducing adjuvant, such as a2,2'-anhy-dropyrimidine, a derivative thereof or auridine phosphorylase inhibitor. Alsoprovided are compositions and kits thatfind use in practicing embodiments ofthe invention. The methods andcompo-sitions find use in a variety ofapplica-tions, including the treatment of avari-ety of different disease conditions.
