Described herein are novel methods and compositions useful in modulating the permeability of a P-glycoprotein expressing barrier of a tissue or an organ, such as the blood- brain barrier, of a subject and improving transport of an agent across the barrier by co¬ administration of the agent with a LPAl Receptor ligand, wherein the LPAl Receptor ligand is administered at a concentration sufficient to reversibly inhibit transport activity of P glycoprotein. Further described herein are compositions and methods useful for treating a condition or disease in a subject by administering to the subject a composition that comprises a therapeutic agent effective against the condition or disease and an LPAl Receptor ligand at a concentration sufficient to reversibly inhibit transport activity of P glycoprotein thereby increasing transport of the therapeutic agent across a P-glycoprotein expressing barrier of a tissue or an organ, such as the blood brain barrier of the subject.
