A method of screening candidate agents for their ability to modulate RAGE activity where such RAGE activity is induced by an active co-located GPCR, the method comprising the steps of: contacting a RAGE polypeptide with a GPCR polypeptide in the presence of a candidate agent where the GPCR polypeptide is constitutively active and/or is activated by addition of an agonist, partial agonist or allosteric modulator of that GPCR； and detecting whether the candidate agent is a modulator of RAGE ligand-independent activation of RAGE by activated co-located GPCR by detecting an effect indicative of modulation of RAGE activation by the presence of the candidate agent and/or by detecting RAGE-dependent signalling that is modulated by the presence of the candidate agent.L'invention concerne un procédé de criblage d'agents candidats pour leur capacité à moduler l'activité RAGE, une telle activité RAGE étant induite par un GPCR co-localisé actif, le procédé comprenant les étapes consistant à : mettre en contact un polypeptide RAGE avec un polypeptide GPCR en présence d'un agent candidat dans lequel le polypeptide GPCR est actif de manière constitutive et/ou est activé par addition d'un agoniste, d'un agoniste partiel ou d'un modulateur allostérique de ce GPCR ； et détecter si l'agent candidat est un modulateur de l'activation de RAGE indépendante du ligand RAGE par un GPCR co-localisé activé par détection d'un effet indicatif de la modulation de l'activation RAGE par la présence de l'agent candidat et/ou par détection d'une signalisation dépendante de RAGE qui est modulée par la présence de l'agent candidat.
