A method for the prediction of male sterility treatment results through determination of fragmentation of DNA. Not damaged and not prepared fresh (not more that 1 h. 30 min after sampling) sample of ejaculate is dissolved in phosphate buffer up to concentration of spermatozoids 5-10 million/ml, after that 25 µl of cellular suspension is applied to object plate treated with agarous micro-gel at a temperature of . Then the plates with preparation are immersed to solution of hydrochloric acid with concentration within 0.1 N and 0.3 N for denaturation of DNA of spermatozoids. After that with doping solution &ndash mixture of solutions of sodium chloride at concentration within and , dithiothreitol at concentration between and , 2-amino-2-(hydroxymethyl)-1,3-propandiol at concentration from to and TritonaX-100 at concentration from 0.5 to 1.5 %, with bringing pH to values 7-7.5, at that most nucleoproteins are removed, at absence of massive DNA damage, nuclei are formed with large circles of DNA loops. Evaluation of the test result is carried out by means of fluorescent microscopy, with calculation of at least 200 spermatozoids for each sample and with calculation of percent of forms with DNA fragmentation, if less than 30 % of cells have fragmentized DNA, this is a positive prognostic factor for occurrence of clinical pregnancy of a woman.Спосіб прогнозування результатів чоловічої неплідності шляхом визначення фрагментації ДНК.
