A microRNA-based short hairpin RNA for gene knockdown of the N-methyl-D-aspartate receptor NR1 subunit, comprises a first fragment, a second fragment and a joining fragment. The second fragment is a palindrome with an inverted repeat sequence of the first fragment. The joining fragment composed by bases at random, and the two ends of the joining fragment connect with the first and second fragment, respectively. The first fragment comprises a dicer-cutting site, and a Drosha-cutting site, and between the two sites is a silencing fragment. The silencing fragment is homologous to the RNA sequence of NMDA receptor NR1 subunit. The microRNA-based short hairpin RNA has a sequence set forth in SEQ ID NO: 5 or 6.
