A method of evaluating the response of a tumor to a DNA-breaking treatment from a sample of cells of said tumor wherein: (a) a cell sample is prepared from cells taken from said tumor; (b) subjecting said cell sample to DNA-disrupting treatment characterized by a D dose, (c) determining on said cell sample the average number of nuclear foci obtained with a pH2AX marker at the observation times t (these numbers means being respectively called NpH2AX (t)), said observation times t being the time t = 0 min (called t0, state before administration of the dose D) and at least one observation time selected from t = t1, t2 , t3 and t4 after dosing D; (d) determining at least one parameter or score for characterizing the response of the sample to said DNA-breaking processing, using at least the average numbers NpH2AX (t), and wherein method • t4 is a fixed value which represents the time for which the rate of DNA breaks reaches its residual value, • t3 is a fixed value which represents the time after which approximately 25% of the double-strand breaks (DSBs) are repaired in control cells from radio-resistant patients, • t2 is a fixed value that represents the time after which approximately 50% of CBDs are repaired in control cells from radiation-resistant patients, • t1 is a fixed value that represents the time after which the number of recognized CBDs reaches its maximum in control cells from radiation-resistant patients.
