Methods and compositions related to the manipulation of proteins having L-cystine / cysteine degrading enzyme activity are described. For example, in certain embodiments, modified cystathionine-γ-lyases can be disclosed that contain one or more amino acid substitutions and are capable of degrading L-cystine / cysteine. Furthermore, certain aspects of the invention provide compositions and methods for the treatment of cancer with L-cystine / cysteine using the disclosed proteins or nucleic acids.
