Methods and compositions relating to modification of proteins having L-cystine / cysteine degrading enzyme activity are described. For example, in certain embodiments, modified cystathionine-γ-lyases can be disclosed that contain one or more amino acid substitutions and that can degrade L-cystine / cysteine. Furthermore, certain aspects of the invention provide compositions and methods for treating cancer with L-cystine / cysteine using the disclosed proteins or nucleic acids.
