A method for producing bioactive substance-encapsulated ethosome comprises the following steps: producing an aqueous bioactive substance solution； producing a lipid-dissolved solution obtained by dissolving lipid in ethanol； stirring the aqueous bioactive substance solution and the lipid-dissolved solution together so as to produce a hydrated liquid crystal phase by which the lipid-dissolved solution is hydrated； and adding purified water to the hydrated liquid crystal phase, and then stirring the same so as to produce an ethosome solution. The ethosome can remarkably improve stability and encapsulation rate of drugs, compared to existing ethosome.생리활성물질이 봉입된 에토좀 제조 방법은 생리활성물질 수용액을 생성하는 단계, 지질을 에탄올에 용해시킨 지질 용해액을 생성하는 단계, 상기 생리활성물질 수용액 및 상기 지질 용해액을 함께 교반하여 지질 용해액이 수화된 수화 액정상을 생성하는 단계 및 상기 수화 액정상에 정제수를 첨가하고 교반하여 에토좀 용액을 생성하는 단계를 포함한다.
