PROBLEM TO BE SOLVED: To provide a method for producing multipotent, pluripotent and self-renewing cells by effective introduction or overexpression of a specified transcription factor that enables the conversion of differentiated cells or somatic cells into multipotent cells, which enhances differentiation into various cell types in the body and enables further differentiation.SOLUTION: Provided is a culture method comprising the steps of: selecting a non-pluripotent somatic cell, into which a protein(s) or a sequence(s) including one or more of long (Prr+)numb isoform, Oct4, Sox2 and Nanog are introduced or overexpressed; growing the selected cells in a growth medium containing a cytokine; and maintaining the cells under the culture conditions until a desired number of the pluripotent cells are obtained. Also provided are a method for gene generation, use of multipotent cell populations, use of pluripotent cell populations, use of nerve cell populations, use of muscle cell populations, and use of other desired cell populations such as the HIV resistant cell populations.【課題】分化された又は体細胞を多能細胞に変換可能にする特定の転写調節因子の有効的導入又は過剰発現により生体内の様々な細胞への分化を促進し、更なる分化を可能にする多能、多能性、自己再生可能な細胞を産生する方法とその使用。【解決手段】非多能性の体細胞を選択し、1つまたはそれ以上の長(Prr+)numbアイソフォーム、Oct4、Sox2、及びNanogを含むたんぱく質又は配列を導入し又は過剰発現させ、サイトカインを含む成長培地において前記選択された細胞を増殖させ、所望の数の多能性細胞が得られるまで、これらの培養条件下で前記細胞を維持する培養方法。遺伝子生成の方法、多能な細胞集団の利用、多能性の細胞集団の利用、神経細胞集団の利用、筋細胞集団の利用、またHIV耐性細胞集団のような他の望まれる細胞集団の利用。【選択図】図1
