PROBLEM TO BE SOLVED: To provide a method for producing a double-stranded polypeptide or protein by recombinant expression in an E. coli host cell.SOLUTION: The method for producing the polypeptide or protein comprises: a step of modifying a polypeptide or a protein in nucleic acid level； a step of introducing the product modified in nucleic acid level into an E. coli cell； a step of culturing and dissolving the host cell； and a step of isolating the double-stranded polypeptide or protein. The polypeptide or protein is a botulinum neurotoxin, especially botulinum neurotoxin type A (BoNT(A)).
