Monoclonal antibodies, or antigen-binding fragments thereof, that bind to ERG,and more specifically, to anepi-tope formed by amino acids 42-66 of ERG3 are disclosed. The monoclonalantibodies can be non-human antibodies (e.g., rabbitor mouse) or humanized monoclonal antibodies having the CDR regions derivedfrom those non- human antibodies. In otherem-bodiments, the monoclonal antibodies are chimeric, having the light andheavy chain variable regions of a non-human ERGanti-body. Methods of using the antibodies to detect ERG, or fusion proteinscomprising all or part of an ERG polypeptide, such as anERG polypeptide encoded by a TMPRSS2/ERG, SLC45A3/ERG, or NDRG1 /ERG fusiontranscript, are also provided, includingmethods of detecting ERG or ERG fusion events in a clinical setting. Theantibodies can also be used to inhibit the activity ofERG or fusion proteins comprising all or part of an ERG polypeptide, such asan ERG polypeptide encoded by a TMPRSS2/ERG,SLC45A3/ERG, or NDRG1 /ERG fusion transcript and to treat malignanciesassociated with overexpression of ERG or an ERGfusion event, such as prostate cancer, Ewing's sarcoma, acute myeloidleukemia, acute T-lymphoblastic leukemia, endothelialcan-cer, and colon cancer.
