PURPOSE: A pharmaceutical composition containing an Acer okamotoanum extract for preventing and treating thrombosis is provided to effectively suppress thrombin and thrombosis, and to prevent and treat thrombosis such as ischemic stroke and hemorrhagic stroke. CONSTITUTION: A method for preparing a thrombin inhibition active material comprises: a step of extracting Acer okamotoanum leaves with water or an organic solvent； a step of fractioning with an organic solvent； and a step of purifying amicon molecules. The organic solvent is selected among alcohol, C1-C4 anhydrous or hydrous alcohol, and a mixture of low class alcohol and water. The thrombin inhibition active material is high molecular polysaccharides with thermal stability and acid stability. A pharmaceutical composition for preventing and treating thrombosis contains an ethyl acetate fraction as an active ingredient. [Reference numerals] (AA) Thrombin time； (BB) Prothrombin time； (CC) APTT time； (DD) Concentration(ug/ml)본 발명은 울릉도 우산고로쇠나무(Acerokamotoanum) 추출물을 유효성분으로 함유하는 트롬빈 저해 혈전증 예방 및 치료용 약학 조성물에 관한 것으로 본 발명의 우산고로쇠 나무 추출물은 물, 주정, 에탄올, 메탄올 등의 다양한 용매로 추출한 후, 상기 추출물을 순차적 유기용매 분획하여 획득되는 것으로 급성경구독성 및 만성독성이 나타나지 않으며, 열 안정성이 우수하고, pH 2의 산성조건 및 혈장내에서도 트롬빈 저해 활성의 손실이 나타나지 않아 이를 유효성분으로 함유하는 약학 조성물은 트롬빈 저해활성을 나타내어 혈전 생성을 효율적으로 억제할 수 있으며, 혈행개선을 통해 허혈성 뇌졸중 및 출혈성 뇌졸중과 같은 혈전증의 예방 및 치료용으로 사용할 수 있으므로 추출액, 분말, 환, 정 등의 다양한 형태로 가공되어 상시 복용 가능한 제형으로 조제할 수 있는 뛰어난 효과가 있다.
