The invention provides a method for the identification and isolation of a gene coding for a HMW glutenin allele variant having a favorable influence on gluten quality. The invention further provides nucleic acid molecules comprising the gene of the allele variant and its derivatives, and the use thereof in developing transgenic wheat varieties with better quality gluten. The invention also relates to methods for creation and identification of mutations improving gluten quality. The invention also concerns the mutant -preferably transgenic- plants produced applying the methods of the invention, and processed derivatives of the plants according to the invention. The claimed invention is useful for developing new wheat varieties with enhanced quality gluten.
