Disclosed is a composition comprising: (a) Iduronate 2-Sulfatase produced by mammalian cells co-expressing the Iduronate 2-Sulfatase and a formylglycine generating enzyme (FGE), and (b) a pharmaceutically acceptable carrier, excipient or diluent wherein the Iduronate 2-Sulfatase has increased ratio percentage of active Iduronate 2- Sulfatase as compared to a control Iduronate 2-Sulfatase produced by the same mammalian cells in the absence of the FGE.
