Bone grafts and constructs comprising stem cells are provided. Exemplary bone grafts include osteogenic stem cells seeded on osteoconductive cortical-cancellous bone and / or osteoinductive demineralized bone scaffolds. An exemplary construct comprises an extracellular matrix on a synthetic scaffold which is secreted from the MSC seeded on a synthetic scaffold. Methods of making the bone graft and scaffold are also provided. There is further provided a method of promoting bone healing and treating wound healing by administering the bone graft and construct to a mammal in need thereof.
