Provided in the present invention is a busulfan injection with a simple formula and stable quality. The injection is prepared by the following method: (1) dissolving busulfan in a mixed solvent of N,N-dimethylacetamide and polyethylene glycol 400 at a volume ratio of (17-57):(83-43) to obtain a busulfan solution with a concentration of 1-7.5 mg/mL and (2) filtering and performing aseptic filling, wherein the temperature of the solution is 4°C-25°C during the dissolution process of step (1), and/or an inert gas is filled until saturated during the operation process. Not only is the stability of the busulfan injection obtained by the preparation method of the present invention improved, but also the safety of the injection is effectively guaranteed after being preserved for a long time.
