Disclosed is a pharmaceutical composition comprising nanoparticles which comprise at least one lipid and RNA encoding at least one antigen, wherein: (i) the charge ratio of positive charges to negative charges in the nanoparticles is between 1.4:1 and 1:8 and/or (ii) the zeta potential of the nanoparticles is 0 or less at physiological pH. Further disclosed is the use of such a composition in the manufacture of a medicament for inducing an immune response and methods of producing said composition.
