A conjugate of polyethylene glycol and a pharmaceutical molecule and a preparation method thereof, and an application of the conjugate in preparation of polyethylene glycol bi-naloxol. The preparation method comprises: (1) preparing a compound of formula I with HO-CH2-(CH2OCH2) n-CH2-OH as a reactant and (2) reacting the pharmaceutical molecule or the pharmaceutical molecule modified by a protecting group with the compound of formula I in a nucleophilic substitution reaction under an alkaline condition to obtain a compound of general formula II, where N is an integer from 0-20, L is a leaving group selected from a group consisting of -OTs、-OMs、-OBs、-OTf、-F、-I、-Br and -Cl, and X1 is a first pharmaceutical molecule or a pharmaceutical molecule group modified by a protecting group.Linvention concerne un conjugué de polyéthylène glycol et une molécule pharmaceutique et un procédé de préparation associé, et une application du conjugué dans la préparation de polyéthylène glycol bi-naloxol.
