Provided herein are synthetic living tissue structures comprising multiplelayers of fibers deposited in solidified formfrom a 3D bioprinter, together with kits and methods of use related thereto.The fibers comprise a plurality of mammalian cellsdispensed within a solidified biocompatible matrix. The structural integrityof the fiber is maintained upon and after deposition withoutany additional crosslinking. The fiber is continuously bioprinted through atleast two layers of the structure. In one aspect, syntheticmuscle tissue structures exhibit a readily assayable contractilefunctionality.
