A recombinant herpes simplex virus type-1 (HSV-1) has been constructed thatcarries a deletion of one of the twoviral .gamma.1 34.5 genes and other immediate early genes, which render thevirus able to selectively replicate in cancer cells but not efficientlyreplicate in normal cells, and in which specific mutations have beenintroduced to enable the virus to spread among cancercells by virus-induced fusion. Specifically, syncytial mutations have beenintroduced in the genes coding for glycoprotein B andglycoprotein K of the virus, enabling high replication and spread of the virusin cancer cells in the presence of substantially loweramounts of .gamma.1 34.5 protein, which is required for optimum infectiousvirus produced and virus-induced cell fusion. Other knownsyncytial mutations could also be introduced that would render the virus ableto cause extensive virus-induced cell fusion. In addition,other known mutations can be introduced to limit the spread of the virus tothe nervous system. These altered viruses or theisolated bacterial chromosomes could be used to treat various cancersincluding breast, liver, colon, and other tissues.
