The present disclosure relates to a method of preparing a solution of factor Xa (fXa) antidote and its lyophilized formulation. Suitable aqueous formulations suitable for lyophilization may include fXa antidote, solubilizer, stabilizer and crystalline components, and the formulation does not disintegrate during lyophilization. The present disclosure provides a lyophilized formulation of a derivative of Factor Xa (fXa) protein, also referred to as “r-Antidot”. Compared to the wild-type fXa protein, r-Antidote has modifications in the Gla domain and active site and retains the ability of fXa to bind to fXa inhibitors, but does not aggregate to form a prothrombinase complex.
