A test device for electrochemical analysis on leukocyte esterase in a test sample is disclosed. The test device comprises an electrode module, an electrochemical control /measurement module, a data acquisition/ storage/ processing module, and a substrate solution. The electrode module including a working electrode, a reference electrode and an auxiliary electrode can provide a volume space as a platform to drop the test sample onto their surfaces. The electrochemical control /measurement module, electrically connected with the electrode module, receives the corresponding electrochemical signals caused by the test samples. The data acquisition/ storage/ processing module, electrically connected with the electrochemical control and measurement module, processes and identifies the received signal change tendency for further calculation. The substrate solution can be catalytically hydrolyzed by the leukocyte esterase containing in the test sample to form an electro-active product, and which would cause a corresponding change in signal. This electric signal change tendency can be further identified, processed and then calculated to obtain the concentration of leukocyte esterase as well as the number of leucocytes in the test sample.
