PURPOSE:To obtain the title enzyme capable of activating biosynthetic pathways of secondary metabolites important to plants by culturing a transformant containing phenylalanine ammonialyase gene of peas in a culture medium and collecting product from cultured mixture. CONSTITUTION:Seeds of Alaskan pea are sown into vermiculite and allowed to stand at 20 deg.C for 7 day in darkness to produce fruit having average size of 7cm and total RNA is isolated from hypocotyl part of the fruit according to an ordinary method and subjected to oligodT cellulose column chromatography to separate mRNA. Cyclic DNA is synthesized using the mRNA as the template and the cDNA is subjected to restriction enzyme treatment and then bound to a phage vector to prepare cDNA library, which is then subjected to cloning by a plaque hybridization method using a probe to select planes positive in phenylalanine ammonialiase gene. Gene is extracted therefrom and the gene is subjected to restriction enzyme treatment and bound to a vector to transform Escherichia coli and the transformant is cultured to produce phenylalanine ammonialyase of peas expressed by the formula.
