The present disclosure relates to an improved method of converting ascomycin to pimecrolimus. Technical ascomycin is chlorinated with triphenylphosphine and N-chlorosuccinimide (NCS) to afford crude pimecrolimus, which is then further purified by HPLC and subsequent crystallization. The method of the present disclosure makes it possible to remove close homologues of pimecrolimus by high pressure liquid chromatography without prior purification of ascomycin starting material.
