Prodrug formulations of insulin and insulin analogs are provided wherein the insulin peptide has been modified by linkage of the carboxy terminus of the B chain to the N-terminus of the A chain via a metastable dipeptide to form an inactive single chain analog. The prodrugs disclosed herein have extended half lives of at least 10 hours, and more typically greater than 24 hours and are converted to the active two chain form at physiological conditions through a non-enzymatic reaction driven by chemical instability.Linvention concerne des formulations de promédicaments dinsuline et danalogues dinsuline, dans lesquelles le peptide dinsuline a été modifié par liaison de lextrémité carboxy de la chaîne B à lextrémité N de la chaîne A par lintermédiaire dun dipeptide métastable pour former un analogue monocaténaire inactif. Les promédicaments décrits dans la description de linvention présentent des demi-vies prolongées dau moins 10 heures, et plus généralement supérieures à 24 heures et sont convertis en une forme bicaténaire active dans des conditions physiologiques par une réaction non enzymatique stimulée par une instabilité chimique.
