PROBLEM TO BE SOLVED: To provide a composition for treating a disease or disorder of the nervous system. Changes in Reelin levels and Reelin signaling alter cognitive function. This can be accomplished by administering to a patient or subject a therapeutically effective amount of a Reelin repeat fragment or a construct formed from a Reelin fragment repeat. Changes in Reelin levels are associated with various neurodegenerative diseases such as fragile X syndrome, William syndrome, Rett syndrome, Down syndrome, Angelman syndrome, autism, ischemia, hypoxia, Alzheimer's disease and schizophrenia, Can be used to treat nerve injury or stroke. Reelin can also be used to alter dendritic spine density, decreased long-term potentiation, decreased synaptic plasticity, and association learning deficits. It has been found that constructs formed from repeat region 3 of full length reelin and repeat region 5 of full length reelin, or repeat region 3 of full length reelin and repeat region 6 of full length reelin are particularly useful.
