Provided are novel human-derived antibodies specific for Fibroblast ActivationProtein (FAP),preferably capable of selectively inhibiting the enzymatic activity of FAP,and chimeric antigenreceptors (CARs) directed against the human FAP antigen as well as methodsrelated thereto. Inaddition, methods of diagnosing and/or monitoring diseases and treatmentsthereof which areassociated with FAP are provided. Assays and kits related to antibodiesspecific for FAP are alsodisclosed. The novel anti-FAP antibodies can be used in pharmaceutical anddiagnosticcompositions for FAP-targeted immunotherapy and diagnostics.
