The present invention relates to an anticancer protein drug conjugate using a repebody, and a preparation method thereof. More specifically, the present invention relates to a repebody-anticancer protein drug conjugate in which a protein drug having an anticancer activity is conjugated to repebody, which exhibits substrate specificity by connecting a leucine-rich repeat (LRR) structure originated from a variable lymphocyte receptor (VLR) as a variable region on the N-terminus of an LRR family protein having an alpha helical capping motif to a preparation method thereof and to a use thereof. The repebody-anticancer protein drug conjugate according to the present invention is a conjugate in which high substrate specificity and excellent tissue and tumor penetration that repebody provides, are combined with a protein drug which is self-assembled and has an anticancer activity. As such, the repebody-anticancer protein drug conjugate shows superior efficacy to the case of using repebody alone, and shows specific selectivity compared to the case of using the anti-cancer protein drug alone. By utilizing these advantages, the present invention combines, with repebody which specifically binds to a protein overexpressed in cancer cells, an anti-cancer protein drug that kills cells, to thereby selectively kill only the cancer cells, and thus can be useful in the treatment of tumors.본 발명은 리피바디(repebody)를 이용한 항암 단백질 약물 복합체 및 그 제조방법에 관한 것으로, 상세하게는 알파 나선형 캡핑 모티프(capping motif)를 가지는 LRR(leucine-rich repeat) 패밀리 단백질의 N-말단에 VLR(variable lymphocyte receptor)에서 기인한 LRR 구조가 가변부위로 연결되어 기질특이성을 나타내는 리피바디에 자기조립되고, 항암 활성을 가지는 단백질 약물이 접합된 리피바디-항암 단백질 약물 복합체(Repebody-anticancer protein drug conjugate)와 이를 제조하는 방법 및 그 용도에 관한 것이다. 본 발명에 따른 리피바디-항암 단백질 약물 복합체는 리피바디가 제공하는 높은 기질특이성과 우수한 조직 및 종양 침투력과, 자기조립 되고, 항암 활성을 가지는 단백질 약물이 결합한 것으로써, 리피바디 단독으로 사용하는 경우에 비해 월등한 효능을 지니고, 항암 단백질 약물 단독으로 사용하는 경우에 비해 특이적 선택성을 지니는 장점을 가지고 있으며, 이를 활용하여 암세포에 과발현되는 단백질에 특이적으로 결합하는 리피바디에, 세포를 사멸시키는 항암 단백질 약물을 결합시켜 암세포만을 선
