The present invention relates to a medium composition for in vitro culture of Polygoni Multiflori Radix and a mass production method of seedling using the same. More specifically, it was confirmed that differentiation of root tubers is excellent by selecting a medium having excellent growth rate through in vitro culture of Polygoni Multiflori Radix using 10 types of media and cultivating the medium using kinetin and IAA or NAA as a plant growth controller. It was also confirmed that culture seedling having excellent differentiation of root tubers can be produced by controlling the content of inorganic salts in the medium and the produced culture seedling makes soil circulated well.본 발명은 하수오 기내배양용 배지 조성물 및 이를 이용하는 종묘 대량 생산방법에 관한 것이다. 상세하게는 10종의 배지를 사용하여 하수오 기내배양하여 생장율이 우수한 배지를 선정하고, 이를 기반으로 식물생장조절제로 키네틴과 IAA 또는 NAA를 이용하여 배양한 것이 덩이뿌리 분화가 우수하다는 것을 확인하였다. 또한 상기 배지 내의 무기염류의 함량을 조절하여 배양함으로써 덩이뿌리 분화가 우수한 배양묘를 생산하였고, 생산된 배양묘가 토양순화가 잘되는 것을 확인하였다.
