An intracellular selection systemallows concurrent screening for peptidebioactivity and stability. Randomizedrecombinant peptides are screenedfor bioactivity in a tightly regulatedexpression system, preferably derivedfrom the wild-type lac operon. Bioactivepeptides thus identified are inherentlyprotease- and peptidase-resistant. Alsoprovided are bioactive peptides stabilizedby a stabilizing group at either theN-terminus, the C-terminus, or both.The stabilizing group can take the formof a small stable protein, such as theRop protein, glutathione sulfotransferase,thioredoxin, maltose binding protein,or glutathione reductase, or one or moreproline residues.
