The present invention provides a novel method for enhancing engraftment of haematopoetic stem cells by an ex-vivo pretreatment comprising the steps of obtaining a sample containing haematopoetic stem cells and admixing a prostacyclin analogue to obtain a mixture, incubating said mixture for a period of time sufficient to stimulate G alphas-signalling in said cells and optionally and isolating said stimulated cells. Further, a composition comprising a prostacyclin analogue for use in the treatment of individuals undergoing haematopoetic stem cell transplantation is provided.Cette invention concerne un nouveau procédé pour améliorer la prise de greffe des cellules souches hématopoïétiques par un prétraitement ex-vivo comprenant les étapes consistant à obtenir un échantillon contenant des cellules souches hématopoïétiques et à y incorporer un analogue de prostacycline pour obtenir un mélange, à incuber ledit mélange pendant une période de temps suffisante pour stimuler la signalisation des G-alphas dans lesdites cellules et éventuellement, à isoler lesdites cellules stimulées. Une composition comprenant un analogue de prostacycline destinée à traiter les sujets devant subir une greffe de cellules souches hématopoïétiques est, en outre, décrite.
