A hypotonic composition containing particles penetrating through the mucus, the particles containing a coating that improves penetration through the mucus, and a therapeutic, prophylactic, diagnostic or nutraceutical agent for introducing into the tissues of the mucous membrane, the composition being hypotonic for the epithelium to which it is delivered so that the composition causes the absorption of water by the epithelium, as detected by the rapid transfer of nanoparticles penetrating the mucus to the epithelial surface, and a more uniform coating of the epithelium lial surface, where they provide prolonged mucosal delivery of the agent. 2. The composition of claim 1, wherein the particles penetrating through the mucus include polymer nanoparticles containing a polymer core and, on its surface, a coating that improves penetration through the mucus, the nanoparticles containing a therapeutic, prophylactic, diagnostic or nutraceutical agent for incorporation into mucosal tissues .3. The composition of claim 2, wherein the mucosal penetration enhancing coating is covalently bonded to the polymer core, where the polymer core is a block copolymer containing one or more blocks of surface-modifying material, or where the polymer core contains one block of coating improving material penetration through mucus covalently bonded to one end of the polymer core. 4.
