Disclosed is a method of making a mouse comprising genetically modifying the mouse to include in its germline: (a) a humanised immunoglobulin heavy chain locus comprising at least one unrearranged human VH gene segment, at least one unrearranged human DH gene segment, and at least one unrearranged human JH gene segment operably linked to a heavy chain constant region gene； (b) a humanised immunoglobulin light chain locus comprising a single rearranged human light chain V/J sequence operably linked to a light chain constant region gene； and (c) an ectopic nucleic acid sequence encoding a mouse ADAM6 protein or ortholog or homolog or functional fragment thereof, wherein the mouse ADAM6 protein or ortholog or homolog or functional fragment thereof is expressed from the ectopic nucleic acid sequence.
