PROBLEM TO BE SOLVED: To provide a method for preparing an amino acid ester of maytansinol. SOLUTION: Maytansinol is reacted with an amino acid N-carboxyanhydride (NCA) in the presence of a base and a desiccant selected from the group consisting of molecular sieve, sodium sulfate, sodium sulfate, calcium sulfate, calcium chloride and magnesium sulfate. Disclosed is a method for preparing an amino acid ester of maytansinol. Also disclosed is an improved method in which a nucleophile is added to the reaction mixture after the reaction of maytansinol with the N-carboxyanhydride of the amino acid is complete.
