in the context of vaccine development, reverse vaccinology, or immunoinformatics, is an approach that integrates different computational methodologies for the search for targets and the design of vaccines. thus, immunoinformatics has been highlighted by allowing the use of programs for the prediction of immunogenic epitopes in silico. this strategy has the potential to pursue targets for vaccine development across the predicted proteome of pathogenic organisms. to date, there is no effective vaccine against visceral leishmaniasis used in vaccination campaigns. Given this scenario, this study proposes the use of immunoinformatics to select epitopes and build chimeric polyepitope vaccines to be tested against visceral leishmaniasis. in the first stage, a t and b cell epitope prediction system was built. this system was validated using experimental data of immunogenic proteins already described in the literature, proving the existence of a correlation and association between the number of epitopes predicted for t / b cells and the reported experimental results. then, two chimeric polyepitope proteins were designed from the approach proposed in immunogenic proteins already written in the literature. such formulated chimeras gave rise to two vaccines, vac-1 (chimera a) and vac-2 (chimera b) both associated with the adjuvant saponin. thus, immunogenicity, immunological memory generation and vaccine efficacy were evaluated in balb / c mice submitted to the immunization protocol and challenge with leishmania infantum promastigotes. these vaccines showed immunogenicity and the ability to induce memory t lymphocytes, in addition to reducing the parasitic load in the spleen. this study allowed to certify and ratify the potential of immunoinformatics as a tool to be used in the development of vaccines against visceral leishmaniasis.no contexto de desenvolvimento de vacinas, a vacinologia reversa, ou imunoinformática, é uma abordagem que integra diferentes metodologias computaci
