Provided herein are polypeptides comprising membrane tethered IL-12. Also provided herein are T cells expressing membrane tethered IL-12. Further provided herein is a method of treating cancer comprising administering T cells expressing membrane tethered IL-12. Also provided is a combination therapy comprising T cells expressing membrane tethered IL-12 and a T cell chemoattractant-inducing chemokine. In addition, methods of activating T cells to express NKG2D and methods of using it for the treatment of cancer are provided.
