Summary of this specification Provided herein are stabilized chimeric Fabs derived from parental chimeric Fabs with lambda light chains. The stabilized chimeric Fabs comprise the immunoglobulin heavy chain polypeptide constructs of the parental chimeric Fab and carry the CH1 sequence and the VH sequence, as well as the V lambda-C kappa chimeric light chain construct. The V lambda sequence of the chimeric light chain construct corresponds to that of the parent chimeric Fab and includes one or more stabilizing amino acid modifications that increase the thermal stability of the stabilized chimeric Fab compared to the parent chimeric Fab. The stabilized Fabs are useful as therapeutic polypeptides, or can be used when constructing antibody constructs in other formats.
