The invention provides a method of producing T-cells for adoptive cell therapy comprising: a) isolating T cells from a mammal and contacting the T-cells in vitro with a TNFa antagonist in an amount effective to suppress the differentiation of the T cells with or without the transduction of a DNA or RNA sequence (e.g., T cell receptor, cytokine, chimeric antigen receptor, or micro-RNA) as part of a T cell adoptive immunotherapy or b) administering to a mammal a TNFa antagonist in an amount effective to suppress the differentiation of the T cells following as part of an immunotherapy regimen including T Cell Adoptive immunotherapy and/or anti Pd-1 and/or anti Pd-L1 antibody treatment.Linvention concerne un procédé de production de lymphocytes T destinés à une thérapie cellulaire adoptive, comprenant les étapes consistant à : a) isoler des lymphocytes T dun mammifère et mettre en contact les lymphocytes T in vitro avec un antagoniste de TNFα en quantité efficace pour supprimer la différenciation des lymphocytes T, avec ou sans la transduction dune séquence dADN ou dARN (par exemple, récepteur de lymphocytes T, cytokine, récepteur dantigène chimère ou micro-ARN) dans le cadre dune immunothérapie adoptive de lymphocytes T ou b) administrer à un mammifère un antagoniste de TNFα en quantité efficace pour supprimer la différenciation des lymphocytes T consécutive à un régime dimmunothérapie comprenant une immunothérapie adoptive de lymphocytes T et/ou un traitement danticorps anti-Pd-1 et/ou anti-Pd-L1.
