Disclosed is an isolated human antibody, or antigen-binding portion thereof, that binds to the p40 subunit of IL-12 and/or IL-23, wherein the antibody, or antigen-binding portion thereof, binds to amino acid residues 15, 17-21, 23 and 58-60 of SEQ ID NO:3 (p40), or within 1-10Å of the amino acid residues wherein the sequence is as disclosed in the specification. Also disclosed is the use of the antibody, or antigen-binding portion thereof in the preparation of a medicament for treating a subject suffering from a disorder in which the activity of IL-12 and/or IL-23 is detrimental.
