Disclosed herein are methods and compositions for the delivery of engineered nucleases and donor molecules into the genome of a cell. Specifically, the methods and compositions described relate to DNA minicircle compositions and the use of DNA minicircles (“MC”) for delivery of nucleic acids (eg, therapeutic nucleic acids) to cells. Cells modified by these DNA MCs are also provided.
