PURPOSE: A microsphere manufactured using a biodegradable polylactic acid derivative microsphere manufacturing method not using the organic solvent is provided to be able to use a needle of thinner thickness by changing the microsphere shape to a constant sphere feasibly and effectively on constant emission and initial burst suppression. CONSTITUTION: A polymeric microsphere manufacturing method comprises a) a step of manufacturing polymer solvent containing solution by mixing solution to polylactic acid derivative aqueous solution of below chemical equation 1 with less than 7000 dalton average molecular weight, b) a step of precipitating the drug containing polymer solution by adding to the aqueous solution of polyvalent metal salt, c) a step of manufacturing micro particle in size of 1-150 micrometers by crushing after drying the precipitated drug containing polymer, d) a step of stirring after suspending the micro particle to polyvinyl alcohol aqueous solution, and e) a step of separating the microsphere produced after coolinig the obtained mixture.유기용매를 사용하지 않는 생분해성 폴리락트산 고분자와 다가 금속을 포함하는 단백질 및 펩타이드 함유 고분자 미립구 제조방법이 개시된다. 상기 제조방법에 의해 제조한 미립구는 표면이 다공성이 없는 일정한 크기의 구형 고분자 미립구로서 상기 제조방법을 통해 초기방출의 억제 및 주사의 편이성 증대가 가능하다.
