Provided herein are methods for preparing monodisperse oligo (ethylene glycol)reagent compositions comprising reacting a halo-terminated oligo (ethyleneglycol) with a hydroxyl-terminated oligo (ethylene glycol)； also providedhereinare small molecule drugs that are chemically modified by covalent attachmentofa water-soluble oligomer obtained from the monodisperse compositions toproduce conjugates which can exhibit a reduced biological membrane crossingrate as compared to the biological membrane crossing rate of the smallmoleculedrug not attached to the water-soluble oligomer.
