PURPOSE: A promoter of gene specifically expressed in rice callus is provided to be very useful in stably mass expressing, particularly, the protein which has the medical or biological activity originated from the eukaryote, in rice callus. CONSTITUTION: A rice callus specific promoter is composed of a base sequence of Sequence no. 1. A recombined plant expression vector includes said promoter. An isolation method of the callus tissue specific promoter comprises a step of manufacturing rice 3' tiling 135k microarray by designing 4 probes of the length of 60 bp which moves by 30 bp from 3'-terminal to the 5' direction of each gene originated from rice and fixing to a microarray; a step of synthesizing cDNA with RNA isolated upon rice tissues as a template and hybridizing to the manufactured microarray; a step of identifying genes expressed specifically in callus than in other tissues based on the hybridization result; and a step of isolating the promoter of the identified genes.본 발명은 벼 캘러스에서 특이적으로 발현하는 유전자의 프로모터, 상기 프로모터를 포함하는 재조합 식물 발현 벡터, 상기 재조합 식물 발현 벡터를 이용한 형질전환 식물체의 제조 방법, 상기 방법에 의해 제조된 형질전환 식물체 및 이의 종자, 벼 3' 타일링 (Tiling) 135k 마이크로어레이를 이용하여 캘러스 조직 특이적 프로모터를 분리하는 방법 및 상기 벼 3' 타일링 (Tiling) 135k 마이크로어레이를 제공한다.
