Disclosed are methods of detecting target analytes using covalent analyte binding moieties in proximity detection assays. Specifically, two proximity detection probes, each comprises an analyte binding moiety and an oligonucleotide moiety, are used for the detection with one analyte binding moiety that covalently binds to the targeted analytes. Further disclosed are methods of coupling an analyte binding moiety with an oligonucleotide moiety to form a proximity detection probe during the detection assays.
