The present invention relates to a method for preparing a dermal substitute with increased biocompatibility and, more specifically, to a method for preparing a dermal substitute comprising the step of treating chloroform and/or trypsin. The dermal substitute manufactured by the method inhibits calcification of the dermal substitute by effectively removing lipid components and immunogenic materials of animal tissue and shows excellent wound healing effects of a transplanted part, thereby being used as a wound covering material, an implant for reinforcing soft tissue, a peritoneal substitute, a meningeal substitute, an adhesion preventing agent, etc.본 발명은 생체 적합성이 증진된 진피대체물의 제조방법으로서,보다 구체적으로는 클로로포름(chloroform) 및/또는 트립신(trypsin)을 처리하는 단계를 포함하는, 진피대체물의 제조방법에 관한 것이다. 본 발명에서, 상기 방법에 의해 제조된 진피대체물은 동물 조직의 지질 성분과 면역원성 물질이 효과적으로 제거됨에 따라 진피대체물의 석회화 현상이 억제되고, 이식 부위의 우수한 창상 치유 효과를 나타냄을 구체적으로 확인하였는바, 창상피복재, 연조직 강화용 이식물, 복막 대체물, 뇌막 대체물, 및 유착 방지제 등으로 유용하게 이용될 수 있다.
