The present invention provides a paper substrate capable of forming alginic acid hydrogel on a surface, and a producing method thereof. The paper substrate of the present invention can be used in co-culturing cells and can be used as a platform capable of forming three-dimensional tissues of an organ level. The paper substrate comprises: (a) paper； (b) a polymer thin film including a copolymer of maleic anhydride and styrene； and (c) a divalent cation and a compound.본 발명은 표면에 알긴산 하이드로젤을 형성할 수 있는 종이 기판과 이의 제조방법, 및 표면에 알긴산 하이드로젤이 형성된 종이 기판과 이의 제조방법을 제공한다. 본 발명의 종이 기판은 세포 공동 배양에도 이용될 수 있을 뿐 아니라, 기관 수준의 3차원 조직을 만들 수 있는 플랫폼으로도 사용 가능하다.
