Hinge antibodies that are selectively activatable in a target cell or tissue to treat a disease of a target cell or tissue are described herein. The hinge antibody comprises a functional antibody, two inhibitory domains and four cleavable linkers. Functional antibodies can treat disease in an activated state and have two light chains and two heavy chains. Each repressive domain contains the hinge domain of an immunoglobulin and consists of two peptide arms. Each cleavable linker comprises a peptide substrate capable of cleavage by a specifically or highly expressed enzyme in a target cell or tissue, wherein one of the peptide arms of the repression domain is attached to the N-terminus of one of the light and heavy chains of the functional antibody .
