The present invention relates to the field of gene engineering biotechnology. Specifically disclosed are a method for designing and preparing a tumor-targeted tumor necrosis factor-related apoptosis ligand variant and a pharmaceutical use thereof. The tumor-targeted tumor necrosis factor-related apoptosis ligand variant is produced from a fusion protein consisting of a CD13 ligand, a linking peptide and a tumor necrosis factor-related apoptosis ligand through clonal construction of an encoding gene of the variant by using a gene engineering method, soluble and recombinant expression, and simple separation and purification. The variant produced by the method for preparing the tumor-targeted tumor necrosis factor-related apoptosis ligand variant is well targeted to the tumor tissues and has a significantly enhanced anti-tumor effect, decreases the protein administration dose required for obtaining the therapeutic effect, increases the bioavailability to increase and reduces the treatment cost, and avoids potential toxic and side effects of the tumor necrosis factor-related apoptosis ligand.
