The present invention relates to a method for producing a high concentration megalocytivirus vaccine. More specifically, the present invention relates to a method for culturing a megalocytivirus by using primary fish larvae culture cells, and a method for producing a vaccine by using the cultured megalocytivirus and a high concentration virus tissue crushed liquid obtained from spleen tissue of infected fish. The method for culturing a megalocytivirus by using primary fish larvae culture cells comprises the steps of: (a) obtaining primary fish larvae culture cells of fish infected by a megalocytivirus (b) inoculating a megalocytivirus in the fish larvae culture cells, and culturing the same and (c) separating the megalocytivirus from the fish larvae culture cells. According to the present invention, a megalocytivirus which is difficult to be cultured in vitro can be cultured at a high concentration, and a high concentration virus of infected fish tissue can be utilized as a vaccine, so a vaccine for preventing megalocytivirus infections in all major culturing fish including rock breams which have been difficult to be applied can be produced. Furthermore, the method for producing a vaccine according to the present invention can be utilized as an alternative vaccine for preventing overall different types of megalocytiviruses which may be domestically introduced in the future.COPYRIGHT KIPO 2016본 발명은 고농도 메갈로사이티바이러스 백신의 제조방법에 관한 것으로, 더욱 자세하게는 (a) 메갈로사이티바이러스에 감염되는 어류의 초대 자어 배양세포를 수득하는 단계 (b) 상기 자어 배양세포에 메갈로사이티바이러스를 접종한 후, 배양하는 단계 (c) 상기 자어 배양세포에서 메갈로사이티바이러스를 분리하는단계를 포함하는 초대 자어 배양세포를 이용한 메갈로사이티바이러스의 배양방법 및 상기 배양된 메갈로사이티바이러스 및 감염된 어류의 비장 조직에서 수득된 고농도 바이러스 조직마쇄액을 이용한 메갈로사이티바이러스를 이용한 백신의 제조방법에 관한 것이다. 본 발명에 따르면, in vitro배양이 어려운 메갈로사이티바이러스의 고농도로 배양할 수 있고 또한 감염된 어류 조직의 고농도 바이러스를 백신으로 활용할 수 있어, 현재까지 적용이 불가능했던 돌돔을 포함한 주요 양식어종 모두에서의 메갈로사이티바이러스 감염증의 예방백신을 제조할 수 있다. 또한, 본 발명에 따른 백신제조 방법은 향후 우리나라에 유입가능성이 있는 다른 type의 메갈로사이티바이러스 모두에 대한 예방 백신 대책으로 활용할 수 있다.
